PERBANDINGAN PREPARAT PERMANEN Pediculus humanus capitis PADA PROSES CLEARING MENGGUNAKAN TOLUENE DAN MINYAK CENGKEH (Syzigium aromaticum)

The process of making permanent preparations includes immersion Pediculus humanus capitis in KOH 10%, dehidrasi, clearing, and mounting. Clearing is a process that has the aim of clearing the tissue preparations so that the insect structure can be seen clearly, clearly, and transparently when observed with a microscope. Toluene has the characteristics of being easily decomposed, slightly soluble in water, sweet and sharp, but flammable, so clove oil (Syizigium aromaticum) is used as a substitute for toluene. The purpose of this study was to determine the quality of permanent preparations of Pediculus humanus capitis in the clearing process using toluene and clove oil (Syizigium aromaticum). This type of research is a comparative test by giving treatment to the subject and object of research. The object used in this study was an adult Pediculus humanus capitis with the same size. The study was conducted at the Parasitology Laboratory of the Health Poltekkes, Ministry of Health, Banten in February 2022. Observations of permanent preparations were carried out by assessing the quality of permanent preparations which included clarity, color quality, and the integrity of the body morphology of Pediculus humanus capitis. The results showed that the celaring treatment with toluene obtained 13 preparations with poor quality, 3 preparations with moderately good quality, and no results were obtained from preparations with good quality. Clearing treatment with clove oil (Syizigium aromaticum) resulted in 3 preparations of poor quality, 5 preparations of moderately good quality, and 8 preparations of good quality. The Mann-Whitnet statistical test showed a significance value of 0.002 with a significance level of 0.05, so it can be concluded that there is a difference in the quality of the permanent preparations of Pediculus humanus capitis in the clearing process using toluene and clove oil (Syizigium aromaticum).
